[TESTING] - Imaging | Fluorescent agent
Description
Test the imaging system's ability (with the excitation LED module) to capture fluorescence levels of a fluorescent agent, perfused in a microfluidic chip. In other words, create a standard/calibration curve.
Notes:
- The agent is fluorescein sodium or any other dye close to GFP's excitation/emission spectrum. You can also try to use its pH dependent fluorescence properties, as explained here.
- Check out this protocol on a fluorescence standard curve with fluorescein in Calibration 3: Fluorescence standard curve - Fluorescein Protocol
- To simulate different levels, you can perform serial dilutions of the agent in water or PBS
- Single-layer chips can be used
- Most likely, you won't saturate the signal so your curve, well, will be a linear line.
